Lab. Research

RESEARCH SUMMARY

In recent years, high-throughput technologies, including microarray and next-generation sequencing, have been utilized to better understand the relationship between genetic alternation and cellular response to explore the complex molecular events. Therefore, my research mainly focus on integrating, analyzing the biological data generated by these high-throughput systems and developing computational methods to investigate important research topics in the fields of clinical research and biological medicine studies. In clinical research, I analyzed NGS data from clinical cancer studies and devoted to discovering biomarkers that could contribute to personalized therapy in the future. I also interested in the radiation induced cellular responses/effects including radiation induced cytotoxicity, apoptosis, and gene expression. Through the radiation biological research, it can provide better understand and improve the strategy of radiation treatment in cancer.

Next-Generation Sequencing (NGS) Related Study

With the advances in Next-Generation Sequencing (NGS), it has been broadly employed to study molecular biology and genetics. With the assistance of an Illumina HiScanSQ Sequencing System that we have established, we not only produce high quality sequencing data , but also setup new pipelines and databases to analyze the big data by cloud computer systems. With close cooperation with multi-discipline research teams involved in clinical medicine, statistics, and computer science, we will equip to conduct comprehensive bioinformatic analyses of NGS data and develop novel computing algorithms for integrated data analyses.

Microarray Data Analysis

In our recent studies, the microarray technology has been comprehensively employed to efficiently identify the predicting biomarkers or different subtypes in human cancer genomics. We have shown the power of using microarray in finding SNPs to predict CCRT response in esophageal cancer patients or identify new biomarkers to predict survival of lung cancer patients. With the advantages of microarray technologies, we therefore highly applied in our studies and also develop new bioinformatics algorithms or tools for data analysis.

Radiation biology related study

Radiotherapy is widely applied in cancer treatment. However, lots of cancers, including lung cancer are less sensitive to radiotherapy and conventional chemotherapy. Previous studies have been reported that ionizing radiation generates reactive oxygen species (ROS) such as superoxide anions, hydrogen peroxide, and hydroxyl radicals, which can be induced DNA damages. In our lab, we have found that two closely human lung cancer cell lines, CL1-5 and CL1-0 had different resistance to different genotoxic agents. Hence, we suggest that unidentified DNA repair defects in CL1-5 may exist and such unknown mechanisms lead to distinct responses of radiation treatment between CL1-0 and CL1-5. Therefore, we would like to perform a general survey of genotoxic agents to understand the DNA repair defects in lung cancer cells, and further elucidate possible mechanisms involved in the repair deficiency.
