細胞面對瞬息萬變的內外在環境刺激必須透過訊息傳遞的過程快速且精準的接收和反應。訊息傳遞的過程是一連串訊息分子,如 protein kinases, adaptor proteins, phospholipids, Ca2+等,有層次、次序及方向性交互作用的結果。因此,改變細胞內訊息傳遞途徑中的任何成員,將會影響到細胞的反應;進而造成生物體生理或病理的變化。
  Focal adhesion kinase, 簡稱 FAK 或 p125FAK,是一個細胞內 tyrosine 的磷酸脢,廣泛的存在原核生物中。早期的研究發現,FAK 在 integrin signaling 的訊息傳遞上扮演非常重要的角色;參與 integrin (一種可與細胞外基質結合的細胞表面上雙複合接受體) 的訊息傳遞反應並調控 integrin 相關的細胞功能,例如細胞生長、細胞移動、細胞附著等。因此,許多的證據也顯示,FAK 的活性主要可以受到 integrin 來調控。
 
 
  在嘗試探討是否 integrin 可以直接的與 FAK 進行交互作而對 FAK 刺激其活性,一直都存在相當的爭議。原因是 FAK 的 carboxyl terminal 區域具有一個 focal targeting sequence (簡稱FAT sequence),負責將 FAK 蛋白專一性的分布到 focal adhesion complex (或稱 focal contacts; 為在細胞表面聚集活化的 integrin 分子其細胞內尾端所形成的聚合體) 當細胞附著時。然而,此一 FAT sequence 並不會與 integrin 的細胞內尾部直接結合,而是透過與 talin 和 paxillin (另外的 focal adhesion complex 蛋白分子) 結合,間接的和 integrin 尾部形成聚合。即使如此,許多的研究包括我們近年來的資料顯示,FAK 經由 FAT sequence 分布到 focal adhesion complex 上並不足以完全活化其活性。其他的因子必須共同參與其活化反應。例如:FAK 的第 397 tyrosine (Tyr397, 為一 autophosphorylation site) 氨基酸位置的磷酸化,產生一個可與含有 SH2 domain (Src homology 2 domain) 的訊息傳遞蛋白結合的結合點。因而可將 FAK 所接受的訊息經由 Src、PI3-kinase、Grb7、PLC-γ 等向下傳遞。若將此一 tyrosine (即 Tyr397) 進行點突變成為 phenylalanine,FAK 的活性便會消失,即使此一突變並不影響到 FAK 的 focal adhesion complex 的分布。另外,我們也發現,FAK 的 amino-terminus,有一區域與其他細胞骨架相關蛋白的氨基酸序列有高度的相似性 (homology) 被稱為 four point one and Ezrin / Radxin / Moesin (FERM) domain, 其具有對 FAK 自我活性調控的功能。若將 FAK 的 FERM domain 進行刪除突變,FAK 的活性便會自發性的增加,不論是在細胞附著或是懸浮的狀態之下。基於上述,對於 FAK 活性的調控機制有必要進一步的釐清。同時,相對於 FAK 的 carboxyl terminus,其 amino terminus 顯少被研究其參與活性調控及細胞功能上的角色。因此,本實驗室將對 FAK 的 amino terminus 在 FAK 本身的活性及功能做進一步的探討;並測試與不同的 integrin 結合及交互作用的能力與角色。
  除此之外,我們也將利用 FAK 細胞或組織專一性的基因剔除鼠探討其在生物體內發育、 生理、及病理的角色。藉此,我們希望可建立 FAK 及其所參與的訊息傳導相關聯的動物模型。提供相關疾病未來治療策略的研究和應用。
 
 
  本實驗室對細胞訊息傳遞路徑的探討也涵蓋近年來在分子植物病理學上相當新穎和重要的基因靜默的現象。對於外來病原的入侵植物寄主,一種內生性的抗病反應可造成寄主植物抵抗病原的成功感染。所利用的機制是受到初步入侵的寄主細胞內可降解及產生病原基因體 (包括 DNA 和 RNA)之衍生物,大小約為 21-25 核苷酸長度的單股 RNA 片段 (又稱 short interference RNAs; 簡稱 siRNAs)。之後,以此 siRNA 對病原基因體有專一性互補的特性,對病原基因體配對形成雙股RNA並進一步在細胞內分解。此一反應也因此可擴大為一連鎖反應,造成系統性抗病。然而,目前我們對於植物基因靜默的生化及分子機轉仍然所之有限。
  植物病原病毒為能達成其成功的侵染寄主植物,相對的也演化發展出對抗基因靜默的機制。某些植物病毒可利用其基因體所轉譯的蛋白分子 (suppressor) 干擾植物內生的基因靜默訊息傳遞路徑。因此,自然演化出的抑制作用,使我們可以利用這些拮抗蛋白分子探索植物內基因靜默的分子機制。同時也可以提供在分子植物病理學上新的致病機轉。
